I don't understand
I understand "I'm going to the rest room" this is a natural expression, however, I don't understand what you mean by "Big one, or small one?" this actually makes no sense.

If you go to the restroom in Japan, you sometimes see the sign that says "big" or "small."

hahaha actually “big one or small one” is a funny sentence in chinglish. Big one is a gentle way of saying poop in Chinese and small one indicates pee. So it means “I'm going to the restroom.” “To poo or to pee?” 【////w////

A little unnatural
If you mean a 小 flush or a 大 flush, then the english equivalent is "number one or number two?"
But you'd only ask someone that if you were VERY close and personal.
