There is not really a big difference, it’s just about the context. When you want to say “Give me the mug on the table” you’ll say “Dă-mi cana DE PE masă” “The mug is on the table” - “Cana este PE masă” We use “pe” when we indicate where is localized the thing and “de pe” when we want someone to give us that specific thing
