"as a matter of fact" is often used as a filler or muletilla. It is often followed by a another thought related to the topic being discussed.
"as a matter of course" usually means "something that happens or will happen naturally as part of some process or chain of events.
Examples:
"My wife is smarter than I am. As a matter of fact, she has two degrees in engineering."
"Mi esposa es más inteligente que yo. De hecho, tiene dos títulos de ingeniería."
"If we replace all our lights with LEDs, the reduced electric bill will happen as a matter of course."
"Si reemplazamos todos los bombillas con LEDs, la factura de electricidad se reducirá naturalmente.
"as a matter of course" is not often used, but it makes you sound smart. 😉
