If someone just says now, then it could have started any time before the present. If someone says, "As of now," then it is a thing that happened very recently.
Examples for both:
Dinosaurs are extinct now.
2NE1 is now disbanded.
As of now, I will start my diet.
Since we ran out of ingredients, we have stopped serving that dish as of now.
