Yes it's used. But it's "spend countless of hours"

Yes. To 'spend countless hours' is used. Also 'spent countless hours' for the past tense. Both are used pretty often.
For the other example mentioned here, I would have to respectfully disagree, saying 'spend countless of hours' would be awkward grammar and would not sound very natural to English speakers.
