Question about English (US)

I can't understand the intent of the author in the sentences down below. I guess it's some kind of jokes but it doesn't really hit me. Can you explain in details? Thank you!

Q: Are two examples(protein powder and dietary supplements) relevant to the context? I know there is a word macho, but how?

2) The impulse to dominate anything—in ourselves or in the world around us—ought to raise some eyebrows. I’ll hold on to my Kleenex, thank you very much.
Q: The Kleenex thing. Why?
