'SeƱor' (gentleman, mister) and 'seƱora' (lady, ma'am, miss) are just plain nouns (sustantivos) in Spanish.
'El', 'la', 'los' and 'las' are definite articles. In English they are all equivalent to the article 'The'. They mean to indicate that you are talking about a specific instance of the noun, not any instance.
In Spanish, however, all words have a grammatical gender, either masculine or femenine, that the definite article has to match. The article also has to match whether the noun is singular (one single unit of the noun) or plural (several units). Thus,
SeƱor = Gentleman
SeƱora = Lady
El seƱor = The gentleman
La seƱora = The lady
Los seƱores = The gentlemen
Las seƱoras = The ladies

The usage of the definite articles also has one more rule that I am aware of, at least with Latin American Spanish, and it has to do with whether or not you are speaking to someone or speaking of someone.
āSeƱor Martinez, por quĆ© estuvo usted en el aula ayer?ā - a person is speaking to Mr Martinez.
El SeƱor Martinez estuvo en el aula ayer. - Mr Martinez is being spoken about.
āSeƱorā is also used for āsir.ā

"SeƱor MartĆnez, por quĆ© *estuvo usted en el aula ayer"
"El SeƱor MartĆnez *estuvo en el aula ayer"
But otherwise a true observation.
