"La" を使うと具体的な教室を意味し、"la" なしで授業中を表します。
大まかに
La clase → ある具体的な教室を指します。
En clase → 授業中。

そのニュアンスは、大きい違いがありませんが、家にいることを強調したい場合に「En casa」と言い、場所であることを強調したい場合に「En la casa」と言います。
→ Voy a casa de mi amigo. // Voy a la casa de mi amigo (友達の家に行く).
どっちも同じ意味です。
