There is no real difference. They both are asking the same thing. which is ' what is your opinion or thoughts on the subject.'
For Example:
The teacher gave us each a copy of the Little Red Riding hood story.
She first asked Mary "What is your takeaway on that story"
Mary said that the wolf is a bad monster because he likes eating everything.
The teacher asked John; "What is your take on it".
John said that his opinion is that the wolf could have been starving to the point he ate what he needed to. He is not a real bad monster.
The only main difference is the way it is spelt or said.
