It's understandable, but it would be better if you wrote, "Speaking of the last day, I'm on my last day of vacation."
If you don't want to use the term 'vacation', you can also write, "Speaking of the last day, I'm on my last day of the holiday."

Speaking of the last day, this is the last day of my holidays, or, speaking if the last day, this is my last day of the holidays. Using my twice sounds odd but you will still be understood! Hope this helps!

A better way to say it would be "Speaking of last days, today is the last day of my holidays."
