Murderer and killer are both people who kill a person or something of significance that is alive. If you really want to be specific, a murderer can be a killer, but the action that a murderer performs has to be illegal. A killer can kill, but can do so legally, like in war.

A killer can be somebody who kills another. Murderer and killer are synonyms. However, an assassination is where an assassin kills a person for political reasons, where the target is usually a person of political importance.
