I have mostly done with this book is bad! Incorrect.

"I am mostly done with this book" is correct.
"I have mostly done WITH ~" doesn't work in English.
Similar phrases are:
"I have mostly done ~" -- such as "I have mostly done nothing this vacation."
or
"I have mostly finished" -- this means the same as "I am mostly done". We use "I am mostly done" more.

The first sentence is grammatically correct.
'done' is an adjective here, not a verb because it follows the reflexive verb 'to be'.
"I am done".
- I am tired.
- She is happy.
- She is disappointed.
The second sentence is incorrect.
You can say (but it sounds awkward) "I have mostly done this book". Here, 'book' is the direct object of 'have done'.
Better to say, "I have mostly finished this book".
