Generally, "learn a lot of Xs" means "learn many different Xs", while "learn X a lot" means "spend a lot of time learning X". The phrase "learn a lot of Xs" can mean "memorise the details of a lot of Xs", like recipes or katas, or "memorise a list of Xs" like words or names.
Neither original phrase is perfect English - you don't really "learn cultures", you learn about them, and if you're talking about Japanese life in general, that would just be "culture", not "cultures". I think "learn a lot about many different Japanese subcultures" and "spend a lot of time learning about Japanese culture" would work better.
