"accused of defamation for the actor" does not quite work ("for" is the wrong preposition there.. it actually suggests that the defamation was done on the actor's behalf or for the actor's benefit (but would be an odd way to say that too)).
You can either say "The woman who was accused of defamation by the actor" (the actor is the one doing the accusing), or you could say "The woman who was accused of defaming the actor" (the actor was the one the defamation was done to).
You also don't really need "of him" after "a big fan", since that's implied already. The rest is pretty good, though, so, for example:
"The woman who was accused of defaming the actor used to be a big fan. Love and hate are two sides of the same coin."

@morph4me "a big fan of his" is actually not grammatically correct. (people do say it sometimes casually, but it is technically wrong)
