Honestly I don't hear much of a difference. They are pretty much synonymous. Perhaps "over the weekend" sounds more modern and natural than "during the weekend". There is another similar phrase: "at the weekend", which I would use. The difference is that "over" or "during" express the total time of the weekend, not just a moment. The kind of sentence you hear in the news is: "over the weekend ten people died in car accidents" or "over the weekend we have had some rain". I don't think "during" would be used as much.
