Generally "main character" is best. However, "protagonist" would be the better term if you are writing about a film for a film class. And you could use "antagonist" in that same paper to describe the villain of the film.

Note: Although "supporting character" is a completely fine term and does have its uses, it's not something that average people in the US tend to say very often. So in a casual conversation I would just use the character's name or the actor's name at that point or else their role (like "the sister, "the husband").
