No hay diferencia de sentido, lo que puede cambiar es lo que quieras enfatizar. Además, es más común omitir los pronombres en español "(Yo) tomo el café..."

Él café lo tomo con leche y sin azúcar / (Yo) tomo el café con leche pero no con azúcar / Yo lo tomo (el café) con leche y sin azúcar.
