To be tired from - when you feel tired due to something and feel you need some rest/a break.
e.g. I'm tired from football training this afternoon.
To be tired of - implies you are sick of doing something, bored of it and don't want to continue doing it.
e.g. I'm tired of studying at my desk all day. I can't wait for summer.
