Depends on the usage, but not really.
“Don’t worry about things.”
“I have no worries.”
Those are both fine, but they are different. They are not truly interchangeable because “no” in the second sentence is quantitative, it means zero worries. But “not” in “don’t” is not quantitative, it is instead giving the inverse of worry, like peace or tranquility.

No, it doesn't sound right. In "don't worry," worry is being used as a negative verb. But in your example, the verb is have, so worry needs to be a noun. "Have no worries" works, but we usually shorten it to just "no worries." :)
