It's a particle of respect you add to your words when asking someone to do something. It originated from a shortening of "po'on" meaning lord, which in itself was a shortening of "panginoon" meaning lord. Today tho, "panginoon" usually refers to lord God. In this sense, "po" can be best translated as "sir" or "ma'am" in english, or perhaps like 先生 in chinese, although you don't call someone directly with it. It's always indirect, since that is more respectful.

"Penge po ng kutsara."
When you're asking for something.
"Excuse me po, nakikiraan po."
When you're saying excuse me to strangers.
"Paumanhin po, di ko po sinasadya."
When you're saying sorry to your boss or anyone who is older or has a higher status.

Without po: anong ginagawa nyo? = what are you guys doing?
with po: anong po ginawa nyo po? = what are you guys doing? *polite*
first audio is without po. second is with the po. Po sound more polite, but it also carries a politeness in speech. that's how I can explain it.

@DJ_ONYX having 2 "po"s in a sentence sounds unnatural. the with po sentences are more natural as either "ano pong ginagawa niyo?" or "anong ginagawa niyo po?"
