Your grill is your face or your teeth. It's an analogy to the air intake grill on the front of a car. "Raise the roof" doesn't make complete sense here. It's when the audience at a concert raise their arms, as though they were trying to lift the ceiling. I think Shrek is repeating it without knowing what it means.

basically he's not trying to be annoying, but this doesn't really mean anything since it's a bunch of outdated slang thrown together -- people would look at you like you were crazy if you talked like that
