There isn't really any emphasis on either, but "I can only cook on Wednesdays." sounds much more natural. Written language and spoken language can both be informal or formal, so there's not much difference on the words. Written may use more shortened words though, like LOL, or LMAO, as in slang. In general, if it's normal to write, it should be normal to say.
