Also do you mind explaining the difference in the use of topic and subject markers?? I'm finding it difficult to know when to use which one.

-이/가 can mean "it's ___ who/what".
For example,
존은 학교에 갔다
= John went to school.
존이 학교에 갔다
= It's John who went to school.
*Notice) sometimes we say like the 2nd one to express the surprise. In that case, it means "Omg, John just went to school! What a surprise!"

And when you mention 2 same subjects in one sentence, it can be written as following:
샐리는 안 간다고 말했다
= Sally said (she) wouldn't go.
샐리가 자기는 안 간다고 말했다.
= Sally said she wouldn't go.
Here, the 2nd sentence mentioned 2 same subjects - 샐리(Sally) and 자기(herself) - while the 1st sentence mentioned the subject only once.

When you're using passive voice, it can be written like this:
이 주제에 대해서는 마리가 준비했다.
= For this topic, Mary is the one who has prepared.
마리는 이 주제에 대해서 준비했다.
= Mary has prepared for this topic.

= My friend is no car.
I'm saying both of your sentences are wrong. They don't make sense at all. You should write:
제 친구는 차가 없어요
or 제 친구는 차를 가지고 있지 않아요.

In your first sentence why use the subject marker on 차 and not the object marker since it's the receiver of the verb 없다

존이 나를 때렸어
= John hit me.
나는 차를 가지고 있지 않아
= I don't have a car
= no car is possessed by me
= I didn't choose any car to possess.

What you're asking is a bit different from the above rule though. To make it simple, I will just give you some examples.
-를 가지고 있지 않아 = -이/가 없어
don't have it = have no ___
나는 연필을 가지고 있지 않아
= 나는 연필이 없어
I don't have a pencil
= I have no pencil.
나는 차를 가지고 있지 않아
= 나는 차가 없어
I don't have a car
= I have no car.
