First off is more informal, usually said in a matter-of-fact way to make a statement or express an opinion.
(First off, I hate waking up early. Second off, I don’t even like my school.)
First is usually used to list things or actions in succession.
(First, I’ll wake up at 6:00 AM. Second, I’ll go to school.)
