What is the difference between "What's your favorite day of the week?" and "What's your favorite day of week" ?Feel free to just provide example sentences.

My problem is in using the definite article "the" before "week."

Ok, but I've seen similar sentences in "American English File 1" English teaching book! Page 35. It reads:
"-What's your favorite time of day? Why? -My favorite time of the day is breakfast."
"-What's your favorite time of day? Why? -My favorite time of the day is breakfast."

That's old English. And it's probably an idiom at this point.
