If Jiro has two or more brothers, you would say: Jiro is the shortest of the brothers/Jiro is the shortest among the brothers.
If Jiro has only one brother, you would say: Jiro is the shorter of the brothers/Jiro is shorter than his brother

Thank you everyone to answer my question. I’ve almost understood that.
But I have another question. When we say “ Taro is the oldest in his family.”, we use “in”, but all you said, in the case of “brother”, you said “of” for the bothers. Why in the case of brother, we should use “of”?
