Why in this sentence (1) does it using the "apostrophe+s" ?
(1) You're a friend of Tim's, aren't you?

Both these sentences are correct. There is no deference between them other than the apostrophe, but even without the apostrophe it changes nothing of the meaning.

There is no reason for the ‘s in the first sentence, it’s just there because native speakers are used to using the ‘s possessive rather than “of” to indicate possessive. This is of course different to Spanish, which only has the “of” phrase and doesn’t have an ‘s equivalent.
There are some times where it helps with understanding though, and has a different meaning. See this article on double possessives:

“A picture of my friend” and “a picture of my friend’s” have quite different meanings.
