They can be used interchangeably in many cases, but there is a little nuance.
Demeanor is about how someone appears to be, and is only really used for people. "They have a very shy demeanor" means "they seem very shy."
Manner is about how someone is or does something, and can be used to describe either a person or an action. "They spoke in a very aggressive manner" means "the way they spoke was very aggressive."
