They're the same, but "Firstly" is grammatically incorrect. It's a slang where people use it for casual English speaking conversations.
Examples:
1. Firstly (incorrect grammar): Firstly, I would like to introduce you to my friend.
2. First of all (correct grammar): First of all, I would like to introduce you to my friend.

They mean the same. "Firstly" isn't slangy or wrong, when used to lead into a sentence. It would be wrong if you were counting (so firstly, secondly, thirdly... don't do that).
