Правильно "лишить покоя". "Лишить покою" - разговорная форма, использовать не рекомендуется.

Лишить покоя- взволноваться обеспокоиться, выйти из равновесия. Покоя так будет правильней.
Покою- стремление к покою

"Лишить покою" - устарелая форма, её можно увидеть в книгах, но использовать не надо. "Лишить покоя" - современный нормативный вариант
