Como? Is how, is the way to explain something, you can say , cómo estás? = how are you?,
Qué?=what?
It's like: What do you want? (Qué quieres?)

"Cómo" it's like when you're asking for the way something is. "How are you?" You can also use it when you're explaining something: "I don't know how was his voice".
"Qué" it's a question with just one answer. "What's that? that's a bird." "What happened? She felt bad."
